The COI gene to identify and determine the limits to species widely used. for this purpose, molecular marker COI gene and ultrastructure first male gonopods was used for the possibility of hidden species among 3 morphotype is shown for this species. therefore sampling from this species were donefrom rocky shores along Bushehr province. then the first male gonopods was separated and photographed by scanning electron microscopy (SEM). for molecular studies samples of DNA extracted by using phenol-chloroform and mitochondrial cytochrome oxidase subunit gene segment (COI) was PCR and sequenced. the results of present study showed that the nucleotide sequence of the mitochondrial gene fragment (COI) is not identical in all samples examined and from these 3 morphotypes, Lydia sp.FCH  species was more different  with other morphotypes and it was placed in a separate clade. ultrastructure study of the apical part of the first male  gonopods  showed  that there are  differences in the apical part of  3 morphtypes. what was obtained from this study showed that It is very important to investigate the first male gonopod  and COI gene in separation of species since, identification of  species  is difficult for many similarities morphology.}, keywords = {Lydia tenax,molecular,COI,ultrastructure,gonopod,Persian Gulf}, title_fa = {شناسایی جنس (Lydia (Brachyuran: Oziidae براساس مطالعات مولکولی و فراساختاری اولین گونوپود جنس نر}, abstract_fa = {جنس Lydia متعلق به زیرشاخه سخت‌پوستان و رده خرچنگ‌های حقیقی می­ باشد. تنها گونه‌ای از آن که در خلیج فارس یافت شده است  Lydia tenax نام دارد. از آن­ جایی ­که در قلمرو دریایی گونه­ های شناسایی نشده به ­وفور وجود دارند روش شناسایی سنتی مبتنی بر ویژگی­ های مورفولوژیک شناسایی آن­ ها را با مشکل مواجه می‌کند. اخیرا ژن COI برای شناسایی و تعیین حد و مرز گونه‌ای کاربرد فراوانی دارد به همین منظور از نشانگر مولکولی ژن COI و هم چنین فراساختار اولین گونوپود جنس نر استفاده شد برای این که احتمال وجود گونه‌های مخفی در میان 3 مورفوتایپ این گونه نشان داده شود. بنابراین از گونه منسوب به Lydia  tenax از سواحل صخره‌ای استان بوشهر نمونه ­برداری انجام شد. سپس اولین گونوپودهای جنس نر، جداسازی شده و با میکروسکوپ الکترونی اسکنینگ (SEM) مورد عکس ­برداری قرار گرفتند. جهت مطالعات مولکولی DNA نمونه­ ها با استفاده از روش فنل-کلروفروم استخراج و قطعه ژنی زیر واحد یک سیتوکروم اکسیداز میتوکندری (COI) تکثیر و توالی­ یابی شد. نتایج مطالعه حاضر نشان داد توالی نوکلئوتیدی این قطعه ژنی میتوکندریایی (COI) در نمونه ­های مورد بررسی یکسان نیست و از این 3 مورفوتایپ نمونه  Lydia sp.FCH با بقیه مورفوتایپ­ ها تفاوت بیش ­تری را نشان داد و در کلادی جداگانه قرار گرفت. مطالعه دقیق فراساختار اولین گونوپود جنس نر در 3 مورفوتایپ، نیز تفاوت ­هایی را در قسمت راسی نشان داد. بررسی حاصل نشان داد، زمانی که تشخیص گونه ­ها به ­دلیل شباهت­ های زیاد مورفولوژی مشکل است، بررسی­ های فراساختار اولین گونوپود جنس نر و شناسایی براساس ژن COI در تفکیک گونه­ ها سودمند خواهد بود. }, keywords_fa = {Lydia tenax,مولکولی,COI,فراساختاری,گونوپود,خلیج فارس}, url = {http://www.aejournal.ir/article_63561.html}, eprint = {http://www.aejournal.ir/article_63561_a084a16c97d24246afbb94152dff0950.pdf} }
