In this study, gene expression changes of IFNγ were evaluated with the addition of Valerian and oregano oil in Arian broilers. The experiment was conducted in a completely randomized design with 7 treatments and 4 replications. Experimental treatments included control, antibiotic, probiotic, Valerian oil and oregano oil at levels of 200 and 400 mg/kg. Body weight traits at 6 weeks, feed rate, feed conversion ratio and IFNγ gene expression were measured. To determine the expression of IFNγ gene, the total RNA was extracted from four hepatocyte repeat lymphocytes of different treatments and, after generating the cDNAs of the samples, the expression of the genes was investigated by Semi-Quantitative RT-PCR. To investigate the effect of experimental treatments on traits and compare the mean of treatments with GLM and Duncan test (p <0.05), SAS software (9) was used. The results were analyzed using GLM software (SAS 9) and the mean of the treatments were compared using Duncan test. The results showed that expression of IFNγ gene was significantly different at different levels of treatment, so that it was the highest at 400 mg/kg and the lowest amount of expression was observed in treatment of oregano oil (200 mg/kg). In general, the results of this study showed that the use of oregano oil in broiler chicken diet improves immune function and decreases mortality rate.}, keywords = {broilers,Gene expression,medicinal plants,performance}, title_fa = {بررسی تاثیر سطوح تغذیه‌ای اسانس سنبل کوهی(Valerian) و مرزن جوش(Oregano) بر بیان ژن اینترفرون گاما در جوجه‌های گوشتی}, abstract_fa = {گیاهان دارویی به­دلیل خواص چندگانه در پیشگیری و درمان بسیاری از بیماری‌های دام و طیور به­جای آنتی­بیوتیک‌ها استفاده زیادی دارد. در این مطالعه تغییرات بیان ژن اینترفرون گاما (IFNg) با افزودن اسانس گیاهان دارویی سنبل کوهی و مرزن جوش در جیره جوجه‌های گوشتی آرین مورد بررسی قرار گرفت. آزمایش در قالب طرح کاملاً تصادفی با 7 تیمار و 4 تکرار انجام شد. تیمارهای آزمایشی شامل شاهد، آنتی‌بیوتیک، پروبیوتیک و اسانس گیاهان دارویی سنبل کوهی و مرزن جوش در سطوح 200 و 400 میلی‌گرم در کیلوگرم بود. صفات میانگین وزن بدن در 6 هفتگی، میزان خوراک مصرفی، ضریب تبدیل غذایی و میزان بیان ژن IFNg اندازه‌گیری شد. برای بررسی بیان ژن، پس از استخراج RNA از بافت کبد جوجه‌ها، cDNA ساخته شد و روش Semi-Quantitative RT-PCR به­کار رفت. برای بررسی تاثیر تیمارهای آزمایشی بر صفات و مقایسه میانگین تیمارها از رویه GLM و آزمون دانکن (0/05>p) در نرم­افزار 9 SAS استفاده شد. نتایج نشان داد که بیان ژن IFNg در سطوح مختلف تیمارها تفاوت معنی‌داری دارد به­طوری‌که در تیمار مرزنجوش(400میلی‌گرم در کیلوگرم)بیش­ترین و در تیمارسنبلکوهی (200میلی‌گرم در کیلوگرم) کم­ترین میزان بیان مشاهده شد. به­طورکلی نتایج این تحقیق نشان داد که استفاده از مرزن جوش در جیره جوجه‌های گوشتی باعث بهبود عملکرد سیستم ایمنی و کاهش میزان تلفات می‌شود. }, keywords_fa = {بیان ژن,جوجه گوشتی,عملکرد,گیاهان دارویی}, url = {http://www.aejournal.ir/article_86195.html}, eprint = {http://www.aejournal.ir/article_86195_4a07cb32d400aa52ccf890df943c0ade.pdf} }
