مطالعه تنوع ژنتیکی جمعیت های مختلف جکوی سنگی تیغه دار (cyrtopodion scabrum) در مناطقی از فلات ایران

چکیده

جکوی سنگی تیغه دار (Cyrtopodion scabrum) سوسماری است که به طور وسیع در جنوب غربی قارۀ آسیا و شمال آفریقا پراکندگی دارد. در ایران این گونه به وفور در بیش تر نقاط کشور وجود داشته و معمولاً به عنوان مارمولک خانگی شناخته می شود. به منظور بررسی وجود یا عدم وجود اختلاف ژنتیکی در بین جمعیت های مختلف،54 نمونه از این سوسمار از 4 استان جمع آوری گردید. پس از استخراج DNA، ژن سیتوکروم b ژنوم میتوکندریایی توسط PCR تکثیر و پس از توالی یابی، توالی ها با نرم افزار Mega 5 و PAUP آنالیز شد. یافته های این تحقیق نشان داد که جمعیت های مختلف این گونه از نظر ژن سیتوکروم b میتوکندریایی یک همگونی را نشان داده به طوری که که میانگین تفاوت در بین این نمونه ها بسیار ناچیز و بین صفر تا 0/1 درصد در تغییر بود. نتایج حاکی از این است که این گونه اخیراً وارد ایران شده و جریان ژنی هنوز بین جمعیت های مختلف آن وجود دارد.

چکیده [English]

The keeled-rock gecko, Cyrtopodion scabrum, is widely distributed in the south west of Asia and northern Africa. In Iran, this species is often found in most parts of the country and commonly known as the house lizard. In order to determine whether or not there is genetic difference between various populations, 45 individuals of this lizard were collected from 5 regions. After extracting DNA, the mitochondrial cytochrome b gene was amplified by PCR and then sequenced. The sequences were then analyzed by Mega 5 and PAUP software. The findings of present study showed that different populations of this taxon display a homology in respect to mitochondrial cytochrome b gene, since the average variation among these specimens was negligible, ranged from 0 to 0.1%.  These results indicate that this taxon has recently been introduced to Iran and a strong gene flow still present within its various populations.
