تاثیر میکروکپسول های حاوی پپتید به عنوان جاذب تغذیه‌ دو گونه ماهی زینتی آنجل (Pterophyllum scalare) و دیسکوس (Symphysodon aequifasciatus)

چکیده

کاربرد موثرتر پروتئین نظیر استفاده از هیدرولیز پروتئینی به ­عنوان جزئی از جیره می‌تواند سبب بهبود کارائی جیره و بهبود شاخص‌های رشد شود. تحقیق حاضر باهدف تغذیه دو گونه ­ماهی زینتی Pterophyllum scalareو Symphysodon aequifasciatus با هیدرولیز پروتئینی بافت ماهی ­کولی با استفاده از عصاره آناناس و پوشش‌ دهی آن با استفاده از آلژینات سدیم 3­ درصد و افزودن به ­میزان 1/5 و 3 درصد به جیره و ارزیابی شاخص‌ های رشد و تغذیه در طی 60 روز انجام شد. برای تعیین سطح بهینه عصاره برای هیدرولیز مقایسه با آنزیم پروتئیناز K مقایسه صورت گرفت و با استفاده از روش بردفورد و SDS PAGE میزان پروتئین هیدرولیز شده بررسی گردید. هیدرولیز تهیه شده با استفاده از آلژینات سدیم پوشش دهی گردید و به جیره فرمول شده با نرم‌ افزاز Winfeed­ اضافه و استفاده شد. نتایج نشان داد پروتئیناز سیستئین موجود در عصاره توانایی بالایی در هیدرولیز پروتئین، معادل 45 درصد دارد. نتایج ارزیابی کپسولاسیون با آلژینات سدیم نیز مشخص کرد که 70 درصد هیدرولیز تولید شده پوشش‌ دهی شده است، در بررسی به ­روش بردفورد میزان جذب قرائت شده معادل 0/649 و غلظت پروتئین معادل 141/64 میکروگرم بر میلی‌ لیتر ثبت شد. بیش ­ترین میزان نرخ رشد ویژه (SGR) در رابطه با ماهیان تغذیه شده با جیره حاوی 3 درصد میکروکپسول به ­ترتیب معادل 2/28 و 2/71 گرم برای آنجل و دیسکس مشاهده شد و این تیمار با سایر گروه‌های آزمایشی دارای اختلاف معنی‌داری بود (0/05>P). هم­ چنین کم ­ترین میزان ضریب تبدیل غذایی نیز در این گروه به­ ترتیب معادل 0/94 و 1/1 درصد محاسبه شد.

Effect of peptide-containing microcapsules as absorbent feeding on two species of Pterophyllum Scalare and Symphysodon Aequifasciatus

چکیده [English]

One of the main aspects of fish rearing is nutrition goals. In this regard,use of protein hydrolyse as of the ingredients compare to the composition of ordinary diets could be effective. The aim of the present study were evaluating the growth performance of two ornamental fish species Pterophyllum Scalare and Symphysodon Aequifasciatus, fed by Vimba vimba tissue hydrolyse, by use of pineapple extract,covered by sodium alginate and add at 1.5 and 3 percent of the diet for 60 days. For modelling the optimum level of pineapple extract for hydrolyzing process, comparison was done by proteinase K activity; qualitative and quantitative evaluations were done through SDS PAGE and Bradford method, respectively. The prepared hydrolyzate covered by 3% sodium alginate and added to formulated diet by WinFeed software. The results reveald that pineapple extract due to protease cysteine content has a significant potential in hydrolyzing, 45% of the protein were hydrolyzed. The results were also shown that 70% of the HP covered by alginate, and Bradford results announced the absorption and protein concentration values equal to 0.649 and 141.64 μg/ml, respectively. The highest SGR and the lowest FCR were computed for groups fed by diet containing 3% of covered HP which were equal to 2.28 and 2.71g and 0.94 and 1.1 for Angel and Discus, respectively(P<0.05).
