اثرات پپتید ضدمیکروبی هپسیدین ماهی آزاد دریای خزر در کنترل باکتری بیماریزای Streptococcus iniae در شرایط درون‎ تنی و بررسی اثر آن بر بیان ژن‎ های سایتوکین در ماهی آزاد دریای خزر (Salmo trutta caspius)

چکیده

هپسیدین ها گروهی از پپتیدهای ضدمیکروبی غنی از سیستئن هستند که در ماهیان علیه باکتری های گرم مثبت و گرم منفی و هم ­چنین ویروسها فعال هستند. هدف از انجام تحقیق حاضر، استفاده از پپتید سنتتیک هپسیدین ماهی آزاد دریای خزر­ (CtHep) برای کنترل باکتری Streptococcus iniae­ در شرایط درون تنی و هم­ چنین مطالعه اثرات تنظیم ایمنی این پپتید در ماهی آزاد دریای خزر می باشد. بچه ماهیان آزاد دریای خزر با میانگین وزنی 20 گرم به ­مقدار 1 میکروگرم به ازای هر گرم ماهی با پپتید سنتتیک CtHep تزریق شدند و سپس در معرض باکتری S. iniae قرار داده گرفتند. بعد از 24 ساعت، مقدار بیان ژن های سایتوکین و میزان بار باکتریایی کل در بافت‎ های کلیه و طحال در تیمارهای آزمایشی و گروه شاهد مورد بررسی قرار گرفت. میزان زنده مانی ماهیان نیز تا روز دهم بررسی شد. بیان ژنهای ­IL-1β ،­IL-6 و ­TNF-α در تیمارهای دریافت­ کننده CtHep به­ طور معنی داری نسبت به گروه شاهد افزایش یافت. به­ علاوه،­ میزان بار باکتریایی کل در بافت های کلیه و طحال در ماهیان آلوده شده به باکتری S. iniaeکه CtHep دریافت کرده بودند به ­طور معنی داری کم ­تر از ماهیان آلوده شده ای بود که CtHep دریافـت نکرده بـودند. درصد زنده مانی در ماهیان آلوده شده به باکتری S. iniae که پپتید ­CtHepدریافت کرده بودند، به ­طور قابل توجهی بیش ­تر از ماهیان آلوده ای بود که پپتید دریافت نکرده بودند. مطالعه اخیر نشان می دهد که پپتید ضدمیکروبی هپسیدین ماهی آزاد دریای خزر پتانسیل استفاده به ­عنوان عامل ضدمیکروبی و هم ­چنین به­ عنوان محرک ایمنی در آبزی پروری را دارد.

Hepcidins are a group of cysteine-rich antimicrobial peptides that are active in fish against gram-positive and gram-negative bacteria as well as viruses. The objective of the present study is to use the synthetic peptide of Caspian trout Hepcidin (CtHep) for controlling in vivo Streptococcus iniae infection and also to study the immunomodulatory effects of CtHep in Caspian trout. Juvenile Caspian trout with average weight of 20 g were injected with synthetic CtHep at a dose of 1 µg/g fish, and then exposed to S. iniae bacteria. After 24 hours, the expression of cytokine genes and total bacterial load were evaluated in kidney and spleen tissues of different groups. Fish survival rate was noticed until 10th day of infection. The expression of IL-1β, IL-6 and TNF-α mRNA in CtHep-receiving group significantly increased compared to the control group. In addition, total bacterial load in the kidney and spleen tissues of infected fish receiving CtHep, was significantly lower than infected fish which did not receive CtHep. Survival rate of infected fish receiving CtHep was considerably higher than that of the infected fish which did not receive CtHep. Our study shows that CtHep has the potential to be used as an antimicrobial agent as well as an immunostimulant in aquaculture industry.
