اثر تغییر سطوح گلوکز و فروکتوز در ترکیب رقیق‎ کننده بر پایه تریس بر فراسنجه ‏های حرکتی اسپرم قوچ نگه داری شده در دمای بدن

چکیده

نگه ­داری منی به ­صورت مایع به ­جای نگه ­داری به­ صورت یخ‏ زده، با توجه به کاهش هزینه ‏های آماده ‏سازی و حمل و نقل مورد استقبال بیش ­تری قرار می‎گیرد. هدف پژوهش حاضر چنین بود که ضمن حفظ اسمولاریته بهینه (330 میلی ‏اسمول) در یک رقیق ‎کننده بر پایه تریس-زرده تخم مرغ (EYT)، تأثیر نوع قند (گلوکز و فروکتوز) و تغییر غلظت قند بر نگه ­داری اسپرم قوچ در شرایط انکوباسیون در دمای بدن (37 درجه سانتی ­گراد) مورد بررسی قرار گیرد. رقیق ‎­کننده ­هایی با نسبت ‏های ترکیبی قند به تریس (30 به 300 و 210 به 100 میلی ‎مولاری قند به تریس) با دو قند مختلف (گلوکز و فروکتوز) ساخته شد و با یک رقیق‎کننده استانداری که برای انجماد اسپرم قوچ به­کار می‏رفت مورد مقایسه قرار گرفت. نتایج نشان داد که بدون توجه به نوع رقیق ‎کننده، فراسنجه‏ های حرکتی اسپرم به ­طور معنی ‏داری بعد از دو ساعت از انکوباسیون کاهش یافت. اثر متقابل تیمار در زمان معنی‏ دار نبود و درصد جنبایی کل و پیش­رونده نیز به ­طور معنی‏ داری تحت تأثیر تیمارها قرار نگرفت. با این­ حال، برخی از فراسنجه ‏های حرکتی نمونه‏ های منی انکوبه شده ازجمله حرکت خطی اسپرم ‏ها (LIN) و شاخص ‏های سرعت حرکت اسپرمی (VAp و VSL) با افزودن غلظت قند در رقیق‏ کننده ‏ها بهبود یافت. نتیجه ‏گیری می ‎شود که افزایش نسبت قند به تریس در رقیق ‎کننده‏ های EYT ممکن است موجب بهبود شاخص ‎های حرکتی اسپرم‏ ها در دمای بدن شود. مقاومت اسپرم‎ ها در دمای بدن می‏ تواند به ­عنوان شاخصی برای باروری گامت های نر در داخل دستگاه تناسلی ماده در نظر گرفته شود.

Storage of semen in the liquid form rather than cryopreservation is becoming popular as preparing and transportation is less expensive in this method. While maintaining the optimal osmolarity in all Tris-Egg yolk based extenders (TEY), the purpose of the present study was to investigate the effect of sugar type (glucose and fructose) and sugar concentration modifications on ram semen kinetics during incubation at body temperature. The semen diluents with different Sugar: Tris ratios (30 mM: 300 mM and 210 mM: 100 mM) were made with two different monosaccharide (glucose and fructose) and were compared with a standard extender used for freezing ram semen. The results showed that regardless of the diluent medium, the mobility parameters of sperm significantly decreased after incubation for two hours at 37°C. There was no significant interaction between storage time and type of extender. Also, the percentage of total motility and progressive motility were not significantly influenced by treatments. However, some of the motility parameters of incubated semen samples, including linearity, curvilinear velocity (VCL) and straight-line velocity (VSL) were improved by enhancing concentration of sugars in diluents. It is concluded that an increase in sugar: Tris ratio in TEY extenders, may improve the kinematic parameters of ram semen at body temperature. Thermoresistance of spermatozoids at body temperature can be considered as an indicator of male gametes fertility inside the female reproductive tract.
