بررسی تأثیر نانو ذرات نقره بر آبشش تاس ماهی ایرانی Acipenser persicus با استفاده از روش HNMR

چکیده

در تحقیق حاضر تغییرات بیوشیمیایی و پروفایل متابولیتی بچه ­ماهی تاس ­ماهی ایرانی Acipenser persicus  که در معرض مستقیم نانوذرات کلوئید نقره طی روزهای مختلف قرار گرفته بود بررسی شد. بدین ­منظور، تعداد 480 قطعه تاس ­ماهی ایرانی در وزن رهاسازی با میانگین وزن کل 0/06± 1/8 گرم در دو دوره حاد 96 ساعته و مزمن 10 روزه در معرض سه غلظت متفاوت 0/001، 0/005 و 0/01 میلی ­گرم در لیتر نانوذرات نقره کلوئیدی به­ همراه تیمار شاهد آب شیرین قرار گرفتند تا تغییرات متابولیت­ های آبشش این ماهیان در شرایط مقابله با نانو ذرات نقره کلوئیدی و با روش متابولومیکس بر پایه HNMR سنجیده شود. مقایسه تیمار 0/01 میلی­ گرم در لیتر نانوذرات نقره نمونه­ برداری شده در روز دهم با سایر تیمارها نشان داد که متابولیت گلیسین و گلوتاتیون بیش ­ترین میزان را داشتند (0/05˂p ). استیل کولین نقش تنظیم یونی داشته و بیش­ ترین میزان آن در تیمار شاهد آب ­شیرین مشاهده شد. میزان متابولیت­ های گلیسین، لوسین و والین در تیمار آب­ شیرین نمونه­ برداری شده در روز دهم نسبت به دیگر تیمارها افزایش معنی ­دار از خود نشان داد. در نتیجه­ گیری کلی، متابولیت گلوتاتیون به ­عنوان نشانگر زیستی و عامل حساس به تغییر میزان نانوذرات نقره در بافت آبشش تاس ­ماهی ایرانی در مطالعه حاضر شناسایی شد.

چکیده [English]

In present study, biochemical alterations and metabolite profile of Iranian sturgeon Acipenser persicus which were directly exposed to colloidal silver nanoparticles during different acute and chronic periods were investigated. A number of 480 pieces of sturgeon at releasing weight with mean total weight of 1.8±0.06 g were exposed to three different concentrations of 0.001, 0.005 and 0.01 mg/L of silver nanoparticles and the alterations of their gill metabolites were investigated in compare with the control treatment using HNMR. The comparison of 0.01 mg/L silver nanoparticles sampled on day 10 with other treatments showed that glycine and glutathione metabolites had the highest value with significant differences (P˂0.05). Acetylcholine had an ionic regulating role and the highest level was observed in freshwater control treatment. The amount of glycine, leucine and valine metabolites in freshwater treatment increased on day 10 compared with the other treatments. Overall, the results showed that glutathione acts as a biomarker and as a sensitive agent to the alterations of the amounts of silver nanoparticles in the gill tissue of Iranian sturgeon.
