توالی یابی ژن سیتوکروم اکسیداز در خط شناسه ژنتیکی ماهیان گورخری (Aphanius dispar) و آفانیوس سوفیه (Aphanius sophiae) جدا شده از آب های گرم در ایران

چکیده

این تحقیق با هدف تعیین خط شناسه ژنتیکی ماهیان گورخری (Aphanius dispar)، آفانیوس سوفیه (Aphanius sophiae) جدا شده از آب ­های گرم در ایران انجام گردیده است. نمونه ­های ماهیان موردنظر از مناطق کولو (شهر اشکنان استان فارس)، چشمه گمبان (شهرستان ارسنجان)، جمع­ آوری شدند. بررسی مولکولی با استفاده از آغازگرهای COI جهت خط شناسه ژنتیکی گونه ­های مورد مطالعه نشان داد که گونه­ A. sophiae جدا شده از چشمه گمبان از لحاظ ژنتیکی بسیار نزدیک به گونه Aphanius anatoliae، Aphanius maeandricus و هم چنین Aphanius sureyanus از کشور ترکیه می­ باشد که خود را به ­صورت کلاستر جداگانه نشان دادند. اگرچه گونه dispar نیز با اختلاف ژنتیکی نزدیک به هفت درصد خود را از این گونه جدا و همراه با گونه sophiae  یک کلاستر خواهری را در همین کلاید ایجاد کرده است. در همین حال دو گونه مورد بحث با اختلاف بسیار بالاتری نسبت به گونه­ های fasciatus و stiassnyae از کشورهای پرتقال و ایتالیا به­ صورت یک کلاید جداگانه قابل توجه می ­باشد. بیش ترین اختلاف ژنتیکی بین گونه fasciatus و sophiae به میزان 21 درصد از کشور آلمان مشاهده گردید و بسیار قابل توجه می ­باشد که با توجه به شکاف عمیق جغرافیایی بین مناطق یاد شده دور از انتظار نبود. استفاده از آغازگرهای مورد استفاده در این تحقیق می­ تواند در شناسایی تنوع ژنتیکی، بارکدینگ و ارتباط منشا ژنتیکی گونه ­های هدف موثر باشد.

چکیده [English]

This research was performed in order to determine of DNA Barcoding of Aphanius dispar and Aphanius sophiae isolated from warm waters in Iran. Samples of the desired fish were collected from Kolo areas (Ashkanan city of Fars province), Gomban spring (Arsanjan city). Molecular study using COI primers for DNA Barcoding of the studied species showed that A. sophiae, isolated from Gomban spring are genetically very close to Aphanius anatoliae, Aphanius maeandricus and Aphanius sureyanus from Turkey, which have shown themselves as separate clusters. However, the dispar species, with a genetic difference of about seven percent, has separated itself from this species and, together with the sophiae species, has created a sister cluster in the same keyboard. At the same time, the two species in question, with a much higher difference than the fasciatus and stiassnyae species in Portugal and Italy, are distinct as a separate clade. The highest genetic difference between the fasciatus and sophiae species was observed in 21% of Germany, which is very significant, was not unexpected due to the deep geographical gap between the mentioned areas. The use of primers used in this research can be effective in identifying genetic diversity, barcoding and the relationship of genetic origin of the target species.
