غربال گری فلور باکتریایی جداسازی شده از دستگاه گوارش مولدین ماهی کپور نقره ای (Hypophthalmichthys molitrix) به عنوان پروبیوتیک

چکیده

این تحقیق با هدف غربالگری هفت گونه از نمونه های باکتریایی دستگاه گوارش مولدین ماهی کپور نقره ای  شامل:  Bacillus subtilis، Entrobacteriaceae، Aeromonas، Corynebacterium jijicum، Staphylococcus، Pseudomonas، Unidentified و Micrococcus lutieus به‌منظور تولید پروبیوتیک انجام گردید. آزمایش فعالیت ­های ضد­باکتریایی در شرایط آزمایشگاهی، با سه روش انتشار در چاهک، انتشار در دیسک و روش متقاطع انجام گرفت. فعالیت ضد­باکتریایی نمونه ­هایی که با هر سه روش بیش ترین فعالیت آنتاگونیستیک را نشان دادند  تحت تاثیر pH ، نمک، دما و زمان مختلف بررسی گردید. اثر­بخشی سوسپانسیون سه گونه کاندید پروبیوتیک و باکتری پاتوژن آئروموناس روی درصد مرگ و میر لارو ماهی کپورنقره ای در شرایط آزمایشگاهی با غلظت های مختلف باکتری­های کاندید (105 ، 107 ، سلول در میلی لیتر) و یک شاهد بدون باکتری در آب ارلن یک لیتری شامل لارو ماهی کپورنقره ای بررسی شد. نتایج نشان داد سه گونه Bacillus subtilis ،Corynebacterium jijicum  و Micrococcus lutieus فعالیت آنتاگونیستی در مقابل پاتوژن­ ها دارند. باسیلوس اثر ممانعت کنندگی معنی‌داری را برای پاتوژن در روش انتشار در دیسک با قطر ناحیه ممانعت کنندگی 20 میلی‌متر نشان داد. افزودن 10 میلی‌لیتر از گونه باسیلوس به پاتوژن آئروموناس جمعیت آن را در 12 ساعت  به حداقل رسانید. رشد مطلوب و تولید ترکیبات ضد باکتریایی نمونه مورد نظر در 8=pH و دمای 37 و 30 درجه سانتی‌گراد دیده شد. درصد بقای بین تیمارها و شاهد،  تیمار 96 ساعته بچه ماهیان با گروه‌های باکتریایی کاندید معنی دار بود (p<0/05). بر اساس نتایج حاصله از این تحقیق، گونه Bacillus subtilis غربال شده می‌تواند به‌عنوان یک پروبیوتیک در نظر گرفته شود.

This study was conducted to screen 7 species of the broodstocks silver carp (Hypophthalmichthys molitrix) digestive tracts bacterial flora including Bacillus subtilis, Enterobacteriaceae, Corynebacterium jijicum, Staphylococcus, Pseudomonas, Micrococcus luteus, Aeromonas, and Unidentified. Antibacterial activities of these candidates bacterial flora were conducted under disc diffusion, well diffusion, and cross streak methods. The tree probiotics candidates (Bacillus, Corynebacterium, and Micrococcus) were shown antibacterial activity against pathogenic bacteria. Candidate Bacillus showed a significant inhibition zone (20 ml) against the pathogen in the disk diffusion method. On the other hand, the addition of 10 ml of Bacillus supernatant to Aeromonas pathogenic species leads to killing them completely at 12 hours. The pH 8 and temperature 30 o was found optimum to grow candidates bacteria and produce antibacterial bio-components. There are significant differences between 8 treatments under different dosages of candidates bacteria and control (without bacteria of beaker included silver carp larva) in fish survival (P<0.05). From this, we can conclude that the candidate Bacillus subtilis maybe can be probiotics for silver carp culture.
