اثر افزودن عصاره آبی رزماری در محیط رقیق کننده بر انجمادپذیری اسپرم قوچ

چکیده

یکی از اثرات منفی انجماد منی دام ­ها افزایش رادیکال ­های آزاد و در نتیجه افزایش پراکسیداسیون  غشای سلولی اسپرم  است که در نهایت سبب کاهش باروری منی پس از یخ ­گشایی می ­گردد. برخی گیاهان با داشتن سطوح بالای آنتی­ اکسیدان ­های طبیعی می ­توانند به کاهش پراکسیداسیون کمک نمایند. هدف از مطالعه حاضر، ارزیابی توانایی عصاره رزماری در محافظت از اسپرم قوچ طی فرایند انجماد-یخ ­گشایی بود. در این مطالعه از 4 رأس قوچ نژاد زندی نمونه­ اسپرم جمع ­آوری شد نمونه ­های منی در هر بار اسپرم­ گیری با هم مخلوط شدند. تیمارهای آزمایشی شامل 5 سطح عصاره رزماری (0، 2/5، 5، 7/5 و 10درصد) بودند. سپس، نمونه ­ها منجمد شده و به مدت 3 هفته در مخزن ازت نگه­ داری شدند. پس از یخ­ گشایی پارامترهای کیفی اسپرم شامل جنبایی کل و حرکت پیش رونده، میزان ناهنجاری­ ها، یکپارچگی غشای اسپرم و میزان تولید MDA اسپرم ارزیابی شدند. نتایج حاصل نشان داد که عصاره رزماری به­ طور معنی­  داری در سطوح 5 و 7/5 درصد سبب بهبود جنبایی کل، حرکت پیش ­رونده، یکپارچگی غشاء و زنده ­مانی شد و میزان ناهنجاری­ های اسپرم و تولید MDA را کاهش داد (0/05>p). با توجه به داده ­های حاصله، افزودن سطوح مشخص عصاره رزماری در رقیق ­کننده، اثر محافظتی در فرایند انجماد- یخ­ گشایی اسپرم قوچ زندی خواهد داشت.

چکیده [English]

One of the freezing semen disadvantages is increasing of free radicals leading to enhanced sperm membrane peroxidation then consequently causes lowered semen fertility after thawing. Some plants bearing high level of natural antioxidants can help to decrease semen peroxidation. The objective of the present study is to evaluate the ability of rosemary extract to protect ram sperm from freeze-thaw damages. In this study, ejaculates were collected from 4 Zandi rams. Semen samples were pooled after each sampling. Experimental treatment included five levels of rosemary aqueous extract (0, 2.5, 5, 7.5 and 10 percent). Then samples were frozen and transferred to liquid nitrogen tanks for three weeks. After thawing sperm quality parameters including total and progressive motility, abnormalities, sperm membrane integrity and MDA production were evaluated. The results showed that rosemary extract significantly improved total and progressive motility, sperm membrane integrity and viability, and decreased sperm abnormalities and MDA production at levels of 5 and 7.5 percent (p<0.05). According to the obtained data, adding specific levels of rosemary extract in the diluent will have a protective effect in the freezing-thawing process of Zandi rams sperm.
