مطالعه اثر سیتوتوکسیک عصاره متانولی از تانتاکل های شقایق موکتی (Stichodactyla haddoni) از خلیج فارس بر روی رده سلول های سرطان کولون و مغز انسان در شرایط آزمایشگاهی

چکیده

شقایق موکتی  Stichodactylahaddoni یک جانور ثابت و ساکن اقیانوس است. زهر شقایق ­های دریایی می­ توانند دارای یک منبع قوی از ترکیبات فعال دارویی باشند. هدفازاینمطالعهبررسیاثرکشندگیسم خامجداشدهازشقایق دریایی S. haddoni  از خلیج فارس بررویمیزانمرگومیررده­ هایسلولی سرطانیمغز و کولونبااستفادهازنمکتترازولیومدرشرایطآزمایشگاهیبود .نمونه­ های شقایق دریایی از جزیره لارک خلیج فارس از جنوب ایران صید شدند. عصاره ­گیری متانولی از تانتاکل­ ها انجام شد. سلول­ های سرطانی با غلظت­ های مختلف زهر خام به ­دست آمده از 0/78-100 میکروگرم به ­صورت رقیق­ سازی در طی زمان 24 ساعت در معرض قرار گرفتند. نتایج آنالیز آماری نشان داد فعالیت ضد سرطانی عصاره رویرده ­های سلول­ های سرطانی  تقریباً در همه دوزها فعالیتی مشابه به­ همدیگر داشتند (0/001>P). زهر خام، مرگ و میر بیش­تری روی سلول­ های سرطانی مغز نسبت به سلول­ های کولون ایجاد کرد. نتیجه 50  IC غلظت ­های مختلف زهر خام بر روی رده ­های سلولی مغز و کولون  به­ ترتیب (6/58 ، 54/ 31 میکروگرم) به ­دست آمد. پس  زهرخامS.haddoniمی­ تواندبه ­عنوانیکعاملشیمیاییوجلوگیری کننده قویرشدبرایرده­ هایسلولیبه ­خصوص سرطان مغز مطرحباشد.

Carpet anemone, Stichodactyla haddoni is an ocean resident sedentary animal. The venom of sea anemones could be a potential source of bioactive pharmaceutical compounds. The main goal of this research was to study the killing effects of the methanol extract tentacles from the Persian Gulf sea anemone, S. haddoni against Brain and colon cell lines in vitro using tetrazolium salt. The specimens were collected from coastal waters of Lark, island Persian Gulf (south of Iran). From tentacles methanol extract was performed. These cells were exposing in presence of different Serial dilution 100 to 0.78 μg of venom extract in during 24 hour. In this study statistical analysis showed that activity was similar to in almost all doses (Pvalue <0.001).The extraction induced more lethality on brain cell line than the other cancer cell lines. IC50 of crude venom against brain and colon cell lines was observed at 6.58, 31.54µg respectively during of 24 hour. Therefore, Stichodactyla crude can be considered as a strong agent of growth inhibitor against cancer cell lines especially brain cancer.
