اثرات سمیت سلولی، همولیز و ضدانعقادی عصاره خاره چسب Siphonaria carbo

چکیده

بررسی کنونی به ­منظور تعیین ویژگی سمیت سلولی عصاره ­های استخراجی از شکم ­پای Siphonaria carbo انجام شد. میزان سمیت عصاره ­های آبی و اتانولی این گونه به­وسیله آزمون سمیت یاخته ­ای با استفاده از آرتمیا انجام شد. هم ­چنین اثرات انعقادی و لیزکنندگی روی گلبول­ های قرمز خون انسان بررسی گردید. نمونه ­ها از صخره­ های سواحل بوشهر جمع­ آوری شد. دو روش عصاره ­گیری از بافت تر و خشک با استفاده از حلال­ های آبی و اتانولی مورد استفاده قرار گرفت. نتایج آزمون سمیت یاخته­ ای نشان ­دهنده این بود که همه عصاره ­ها (به ­جز عصاره آبی خشک) اثر کشندگی داشتند و میزان سمیت با افزایش غلظت عصاره همبستگی داشت. عصاره آبی تر بیش­ ترین میزان سمیت (5/98= LC50-24hr) را در مقایسه با سایر عصاره­ ها داشت. نتایج به ­دست آمده از تست همولیز خونی نشان داد اثر همولیتیک معنی­ داری در عصاره ­های استخراجی وجود ندارد. هم­ چنین آزمون ­های انعقادی نشان دادند خاصیت ضدانعقادی در عصاره­ های استخراجی وجود ندارد. به­ طورکلی در بررسی کنونی خاصیت ضدانعقادی قوی مشاهده نشد، ولی بالا بودن اثر کشندگی سلولی عصاره­ ها، نشان ­دهنده احتمال بالای سایر اثرات مانند اثرات ضد توموری و ضد انگلی می ­باشد. روش ­های تخلیص عصاره­ ها به­ منظور دستیابی به کاربردهای دارویی عصاره پیشنهاد می­ گردد.

The present study was conducted to study cytotoxicity of extractions from Siphonaria carbo limpet. Brine shrimp lethality assay, hemolytic effects on erythrocytes and coagulation tests was performed using aqueous and ethanolic extracts. Sampling was performed from Bushehr coastal rocks. Extraction was carried out using wet and dry tissue. Results of toxicity assay on Artemia nauplii showed lethality effects of extracts, except for dried aqueous extract. The lethality of extracts was in correlation to concentration. Wet aqueous extract showed the highest toxicity on tested cells (LC50= 5.98 mg/ml) in contrast to other extracts. The results of hemolysis test revealed no significant hemolytic activity of extracts. The results of anticoagulant assays did not confirm any anticoagulant effects in any of extracts. In conclusion, in spite of the negative results of hemolytic and anticoagulant effects, the cytotoxicity of the extracts might be utilized for further anticancer and antiparasitic screening.
