تاثیر غلظت های تحت کشنده دیازینون بر بافت تخمدان در ماهی گورخری (Danio rerio)

چکیده

سم دیازینون، یکی از سمومی است که به‌عنوان آفت­ کش در باغات و زمین­ های کشاورزی کشور به ­ویژه در منطقه استان گلستان استفاده می­ گردد. با این­ که بوم‌ سازگان‌های آبی محل و هدف استفاده این سموم نیستند، اما مطالعات پایشی شواهدی از حضور آن‌ها و متابولیت ­های آن‌ها را در محیط‌ های آبی ب ه‌ویژه آب‌ های سطحی را نشان می‌ دهد. اثرات دوزهای تحت­ کشنده سم دیازینون تجاری بر آسیب بافت تخمدان ماهی گورخری نابالغ در مواجهه 30 روزه مورد بررسی قرار گرفت. برای این منظور 240 ماهی ماده با میانگین وزنی 0/05±0/2 در 3 تیمار تحت­ کشنده ۵، ۱۰ و ۲۰ درصد از 50 LC سم دیازینون به ­ترتیب  0/8، 1/6، و 3/2 میلی ­گرم بر لیتر و تیمار شاهد قرار گرفتند. در پایان دوره، نمونه ­برداری از بافت گناد ماهیان انجام و مقاطع بافتی تهیه شد. نتایج نشان داد که در گروه شاهد، بافت تخمدان طبیعی و عمده اووسیت ­ها در ابتدا و انتهای مرحله چهارم رسیدگی جنسی بودند.  در تیمار0/8 میلی­ گرم بر لیتر اووسیت­ ها درمرحله سوم و انتهای مرحله سوم رسیدگی جنسی بوده و تعدادی اووسیت در مرحله دوم مشاهده شد. در تیمار 1/6 و 3/2 میلی­ گرم بر لیتر، رشد ناچیز تخمدان مشاهده و اووسیت ­ها عمدتاً در مرحله دوم رسیدگی جنسی قرار داشتند. در این تیمارها هایپرپلازیا در لایه گرانولوزا و فولیکول­های آترتیک مشاهده شد. این روند رسیدگی نشان ­دهنده کاهش رشد اووسیت با افزایش دوز سم می ­باشد. اندازه تخمدان در تیمارهای مختلف به ­ویژه با دوز بالا (3/2 میلی­ گرم بر لیتر) کاهش چشمگیری نسبت به گروه شاهد نشان داد.

Diazinon is one of the pesticides used in gardens and agricultural lands. Although aquatic ecosystems are not the purpose of these toxins, the studies show evidence of their presence and metabolites in aquatic environments, especially surface waters. The effects of sub-lethal doses of diazinon in zebra fish ovary tissue were investigated in 30-day exposure. For this purpose, 240 females with mean weight 0.2± 0.05 were subjected to 5, 10 and 20% LC50, 0, 0.8, 1.6 and 3.2 mg / L,  respectively,  diazinon. At the end trail, samplings were done from gonad tissue and the tissue sections were prepared. The results showed that in the control group, ovary tissue was normal and major of the oocytes were sexually at the beginning and the end of the fourth stage. In 0.8 mg / L, oocytes were in the third and the end of the third stage and a number of oocytes were observed in the second stage. In 1.6 and 3.2 mg / L, a slight increase in ovaries growth was observed, oocytes were in the second stage and hyperplasia in the granulosa layer and atrotic follicles was observed. In general, gonad tissue damage increased with increase in diazinon dose. . This indicates that oocyte growth is declining with increasing dose of diazinon. Ovarian size was significantly reduced, especially in high dose (3.2 mg / l),  compared to the control group.
