اثرات مورفین سولفات بر سطح گلوتامات هیپوتالاموسی و غلظت سرمی هورمون LH در موش های صحرایی نر

چکیده

مورفین از طریق تاثیر بر فعالیت مسیرهای نورونی واسطه ­ای مختلف داخل هیپوتالاموسی اثرات مهاری بر ترشح هورمون­ LH اعمال می­ کند. در تحقیق حاضر، اثرات مورفین سولفات بر ترشح LH از طریق مسیر گلوتاماترژیک در موش ­های صحرایی نر بررسی گردید. بیست موش صحرایی نر بالغ نژاد ویستار به وزن 250-200 گرم در چهار گروه (در هر گروه 5=n) قرار گرفته، گروه ­های 1 و 2 سالین و گروه ­های 3 و 4 یک میکروگرم مورفین را از طریق تزریق درون بطنی مغزی دریافت کردند. در گروه ­های 1 و 3 نمونه­ های خونی قبل از تزریق و 30 دقیقه بعد از تزریق سالین یا مورفین جمع ­آوری و نمونه­ های هیپوتالاموسی جدا گردید. در گروه­ های 2 و 4 نمونه­ های خونی قبل از تزریق و 60 دقیقه بعد از تزریق سالین یا مورفین جمع­ آوری و نمونه­ های هیپوتالاموسی جدا گردید. غلظت سرمی هورمون LH با روش رادیوایمونواسی (RIA) و سطوح گلوتامات هیپوتالاموسی با استفاده از کروماتوگرافی مایع با کارایی بالا (HPLC) اندازه­ گیری شد. مورفین میانگین غلظت سرمی LH را در 30 یا 60 دقیقه بعد از تزریق در مقایسه با قبل از تزریق کاهش داد که این میزان کاهش تنها در گروه 60 دقیقه از نظر آماری معنی ­دار بود. هم­ چنین، با تزریق مورفین میانگین غلظت گلوتامات هیپوتالاموس در 30 دقیقه بعد از تزریق در مقایسه با گروه 30 دقیقه بعد از تزریق سالین از نظر آماری به ­طور معنی ­داری کاهش یافت. کاهش فعالیت نورون­ های گلوتاماتی هیپوتالاموس ممکن است در مسیرهای عصبی کنترل­ کننده اثرات مورفین بر ترشح LH دخالت داشته باشد.

Morphine exerts inhibitory effects on GnRH/ LH hormones secretion via the effects on the activity of different intra hypothalamic interneurons. In the present study, the effects of morphine sulphate were investigated on LH secretion via monoamine glutamate in male rats. Twenty adult male Wistar rats weighing 200- 250g in two group (n=10 in each group) received saline or one microgram morphine via third cerebral ventricle. Then each group divided in two groups. Blood samples were collected before injections and at 30 or 60 min after saline or morphine injections. Hypothalamic samples were dissected 30 or 60 minutes after injections. Serum LH hormone concentration was measured by the radioimmunoassay method and hypothalamic glutamate levels was measured by high performance liquid chromatography (HPLC). Morphine decreased mean serum LH concentration at 30 or 60 min after injections compared to before injections of morphine however this decrease was statistically significant only at 60min group. Also, morphine significantly decreased mean hypothalamic glutamate levels at 30 min after injections compared to 30 min after injections of saline group. Decrease of hypothalamic glutamate neurons activity may be involved in the morphine neural pathways controlling the inhibitory effects of morphine on LH secretion.
