پراکسیداسیون لیپیدهای غشایی و کیفیت پارامترهای اسپرمی در موش های نر نژاد Balb/c تیمار شده با غلظت های مختلف سم دیازینون

چکیده

با توجه به نقش احتمالی دیازینون (DZN) در القاء تولید رادیکال­ های آزاد، احتمال می ­رود که سبب افزایش اکسیداسیون لیپیدهای غشایی اسپرم و در پی آن کاهش کیفیت پارامترهای اسپرم گردد. در این مطالعه اثرات سمی دیازینون بر پارامترهای اسپرم و اکسیداسیون لیپیدهای غشایی اسپرم در مایع سمینال موش نر مورد بررسی قرار گرفته است. در این مطالعه تجربی، 24 موش نر بالغ از نژاد سوری به ­صورت تصادفی به سه گروه تجربی و یک گروه شاهد تقسیم شدند. سم دیازینون با غلظت­ های 5 (گروه A)،  ده (گروه B) و 25 (گروه C) میلی­ گرم به­ ازای هر کیلوگرم از وزن در هر روز، به ­ترتیب به گروه­ های اول تا سوم مورد مطالعه و به ­صورت دهانی به­ مدت پنج هفته داده شد. سپس غلظت مالون دی آلدئید (MDA) در سرم و مایع سمینال به ­روش (TBAR (Thiobarbituric acid reaction  با دستگاه اسپکتروفتومتر مورد آماده ­سازی قرار گرفت. پارامترهای اسپرمی نیز به ­روش ­های معمول میکروسکوپی مورد آنالیز قرار گرفت. کاهش معنی ­داری در میانگین کیفیت پارامترهای اسپرمی در گروه ­های تیمار شده با سم دیازینون، به ­خصوص گروه­ های B و C در مقایسه با گروه شاهد مشاهده گردید (0/001>p). تفاوت معنی­ داری در میانگین غلظت MDA در سرم و مایع سمینال بین گروه­ های تیمار شده A، B، C و گروه شاهد مشاهده گردید (0/001>p).  سم دیازینون با افزایش اکسیداسیون لیپیدهای غشایی سبب کاهش پارامترهای اسپرمی می ­گردد که اثرات آن وابسته دوز نیز می ­باشد.

Regarding the possible role of Diazinon (DZN) to induce free radicals production, it is assumed that it can lead to increased level of lipid peroxidation and then poor sperm quality. In this research, we aimed to investigate the toxic effects of DZN on sperm parameters quality as well as lipid peroxidation level in sera and seminal plasma of male rats. In this experimental research, 24 male rats (Balb/c) were randomly divided into four groups including: controls (n=6), treatment group A (n=6), treatment group B (n=6) and treatment group C (n=6). Commercial DZN was injected orally in a single dose (A=5mg/kg, B=10mg/kg and C=25mg/kg per day). After 5 weeks, animals were sacrificed for Malondialdehyde (MDA) and parameters quality examinations using Thiobarbituric acid reaction (TBAR) and microscopic techniques, respectively.  There was a significant reduction in sperm parameters quality in treatment groups, especially in group C and group B, when compared with controls (p<0.001). There was a significant difference for serum and seminal plasma MDA mean levels between study groups of A, B, C and control groups (p<0.001).  DZN leads to poor sperm quality via increasing of serum and seminal lipid peroxidation levels in a dose dependent manner.
