اثر غلظت های تحت کشنده نانوذرات نقره و نیترات نقره بر بیان ژن HSP70 و آسیب های بافت های آبشش، کبد و روده در ماهی کپورمعمولی (Cyprinus carpio L)

چکیده

در این مطالعه به­منظور بررسی اثرات نقره بر ماهی کپورمعمولی، ماهیان در معرض غلظت­های تحت حاد نانوذرات نقره و نیترات نقره قرار گرفتند. برای این منظور ابتدا غلظت حاد 50% و 96 ساعته برای نانوذرات نقره و نیترات نقره به­دست آمد و سپس ماهیان به­مدت 21 روز در معرض غلظت­های تحت حاد قرار (04/0 و 08/0 میلی­گرم در لیتر نانوذرات نقره و 025/0 و 045/0 میلی­گرم در لیتر نیترات نقره) گرفتند و شاخص­های مورد نظر اندازه­گیری قرار گرفت. پس از درمعرض قرارگیری با دو شکل نقره، عوارض مختلفی در بافت­ها مشاهده گردید از مهم­ترین آن­ها می­شود به برخواستگی اپی­تلیالی، کوتاهی و ریختگی لاملاهای ثانویه آبشش، تخریب و نکروز شدن هسته و هایپرتروف شدن هپاتوسیت­های کبد و هم­چنین تخریب ساختار پرزهای روده و تورم آن­ها اشاره کرد. نتایج این مطالعه نشان داد که در معرض­گذاری بلندمدت با نانوذرات نقره باعث افزایش معنی­دار بیان ژن Hsp70 شد و پس از در معرض قرارگیری ماهیان کپورمعمولی با نیترات نقره نیز، ژن Hsp70 به­شدت افزایش پیدا کرد اما اختلافی بین دو ژن از نظر بیان ژن مشاهده نگردید. هم­چنین بیش­ترین تاثیرات مشاهده شده در بین بافت­های بررسی شده در این مطالعه، در کبد ماهیان در معرض قرار گرفته با دو شکل نقره و در بالاترین غلظت مشاهده گردید و اختلافی از نظر سم­شناسی بین دو شکل نقره مشاهده نگردید.
