بهبود فراسنجه های کیفی اسپرم منجمد-یخ گشایی شده قوچ با افزودن عصاره نعناع فلفلی در رقیق کننده

چکیده

هدفاینپژوهشبررسیتأثیر عصارهگیاهنعناع فلفلیبه ­عنوان یکآنتی­ اکسیدانطبیعیبرکیفیتاسپرممنجمد-یخ­گشاییشدهقوچبود. برای این منظور از چهار رأس قوچ مغانی هفته ‏ای دو بار اسپرم‏ گیری شد و انزال­ ها به­ طور یکسان باهم مخلوط شدند. سپس نمونه‏ ها با رقیق‏ کننده حاوی سطوح مختلف عصاره نعناع فلفلی (0، 2، 4، 8، 12 و 16 میلی ­لیتر در دسی ­لیتر محلول رقیق­ کننده) رقیق شدند. نمونه ­هاپسازطیمراحلسردسازیو انجمادتازمانارزیابیدرداخلازتمایعنگه­ داریشدند. بعد از یخ‏ گشایی، پارامترهای تحرک به ­وسیله سیستم کاسا، زنده‏ مانی به ­روش رنگ‏ آمیزی ائوزین-نیگروزین و تست یکپارچگی غشایی با محلول هیپواسموتیک مورد بررسی قرار گرفت. نتایج نشان داد که افزودن عصاره در سطوح 4 و 8 میلی ­لیتر، درصد تحرک کلی اسپرم ­ها را افزایش داد (0/05<P). هم­ چنین غلظت 4 میلی­ لیتر عصاره به­ طور معنی ­داری تحرک پیش ­رونده اسپرم ­ها را نسبت به گروه شاهد و گروه ­های تیماری حاوی سطوح بالاتر بهبود داد (0/05<P). در رقیق­ کننده حاوی 8 میلی ­لیتر عصاره، درصد صفات LIN، STR و VSL بالاتر بود (0/05<P). هم چنین افزودن سطوح 4 و 8 میلی ­لیتر عصاره، پارامترهای VCL و VAP را بهبود داد (0/05<P). سطوح 4 و 8 میلی ­لیتر عصاره، درصد زنده­ مانی ویکپارچگی غشای پلاسمایی اسپرم ­ها را نسبت به گروه شاهد بهبود داد (0/05<P)، اما ریختشناسیاسپرم­ ها تحت تاثیر تیمارها قرار نگرفت. در نتیجه استفاده از عصاره نعناع فلفلی در رقیق‏ کننده منی منجر به بهبود کیفیت اسپرم منجمد-یخ­ گشایی شده قوچ شد که این اثر وابسته به دوز بود.

چکیده [English]

The aim of current study was to evaluate the effect of Mentha piperita extract as a natural antioxidant on quality of frozen-thawed ram sperm. In this study, four Moghani ram were used for semen collection twice a week. Then semen samples were pooled and diluted with extenders supplemented with different concentrations of Mentha piperita extracts (0, 2, 4, 8, 12 and 16 mL per dL diluent solutions). Following cooling and freezing, semen samples were stored in liquid nitrogen until evaluation. After thawing, the sperm dynamic parameters were evaluated using CASA system. The results showed that the addition of 4 and 8ml/dl extracts resulted in higher (p < 0.05) percentages of total motility compared to control. Addition of 4 ml/dl extract improved progressive motility compared to control group and higher levels of extract groups (P <0.05). The percentages of LIN, STR and VSL were higher (P<0.05) in the extender containing 8 mL/dL extract. Addition of 4 and 8 mL/dL extract to the extender significantly (P<0.05) improved VCL and VAP parameters. Addition of 4 and 8ml/dl extract improved the percentage of viability and sperm plasma membrane integrity compared to control (P <0.05). Sperm morphology was not affected by treatments. In conclusion, supplementation of extender with Mentha piperita L. extract improves post-thawed ram sperm quality in a dose dependent manner.
