بررسی امکان رفع چسبندگی تخم به وسیله آنزیم آلکالاز، اسیدتانیک و سدیم کلراید به جای محلول کاربامید در فرآیند تکثیر مصنوعی ماهی کپورمعمولی (Cyprinus carpio)

چکیده

تخم­ های بسیاری از ماهیان آب­ شیرین بعد از فعال­ سازی و تماس با آب حالت چسبنده پیدا می ­کنند که علت آن وجود مولکول ­های گلیکوپروتئینی در بخش خارجی آن­ هاست. چسبندگی تخم ­ها تحت شرایط تفریخ­گاهی مطلوب نبوده و انباشتگی تخم­ های متصل به ­هم منجربه تلفات دراثر آنوکسی و رشد قارچ­ ها می ­شود. بنابراین یکی از مراحل تکثیرمصنوعی این قبیل ماهیان ازجمله کپورمعمولی (Cyprinus carpio)رفع چسبندگی تخم ­ها پیش از انکوباسیون می ­باشد. در این تحقیق به­ منظور رفع چسبندگی تخم­ های لقاح یافته ماهی کپورمعمولی (Cyprinus carpio) از سه ماده آنزیم آلکالاز، اسیدتانیک و سدیم کلراید استفاده شد و نتایج مربوط به درصد لقاح و نرخ تفریخ با روش متدوال کارگاه (شست­ و شوی تخم­ ها با محلول کاربامید) مقایسه شد. 9غلظت از سدیم کلراید 25، 50، 100، 250، 500، 1000، 2000، 4000 و 10000 میلی­ گرم در لیتر در سه زمان 15، 30 و 60 دقیقه به ­کار گرفته شد. 3 غلظت از اسیدتانیک 500، 1000 و 1500 میلی­ گرم در لیتر در سه زمان 0/5، 2 و 5 دقیقه به ­کار گرفته شد. هر سه غلظت در سه تکرار انجام ­شد. 20 دقیقه پس از فعال­سازی 50 میلی­لیتر آنزیم با غلظت 2 میلی ­لیتر بر لیتر اضافه شده و در زمان­ های 45، 60، 75 و 90 ثانیه در معرض قرار گرفتند. در نهایت 4 بار با آب هچری شست­شو داده شده و به انکوباسیون انتقال داده شدند. نتایج در مورد رفع چسبندگی تخم­ ها به­ وسیله تیمار اسیدتانیک نشان داد که بهترین درصد لقاح (99/57) و نرخ تفریخ (99/46) مربوط به غلظت 500 میلی­ گرم در لیتر در مدت زمان 0/5 دقیقه بود. در مورد رفع چسبندگی تخم­ ها به ­وسیله تیمار سدیم کلراید نشان داد که بهترین درصد لقاح (87/1) و نرخ تفریخ (86/28) مرتبط به غلظت 1000 میلی­ گرم در لیتر در مدت زمان 30 دقیقه بود. بیش ­ترین درصد لقاح (97/52) و تفریخ (96/72) با آنزیم آلکالاز مربوط به تیمار با غلظت 50 میلی­ گرم در لیتر در مدت زمان 45 ثانیه بود که به ­لحاظ آماری اختلاف معنی­ داری با گروه شاهد داشتند (0/05>P).

In this research, in order to eliminate the adhesion of fertilized eggs, common carp of common carp from three materials of alkalase, tannic acid and sodium chloride was used. The results of fertilization rate and hatching rate were compared with the general method of the workshop (egg washing with carbamide solution). 9 concentrations of sodium chloride were used at 25, 50, 100, 250, 500, 1000, 2000, 4000 and 10000 mg / L for three times 15, 30 and 60 minutes. Three concentrations of tannic acid (500, 1000 and 1500 mg / L) were used at three times 0.5, 2 and 5 minutes. All three concentrations were performed in three replicates. After activation, 50 milliliters of the enzyme were added at a concentration of 2 milli liters per liter for 20 minutes and exposed at 45, 60, 75, and 90 seconds. Finally, they were washed 4 times with hatchery juice and transferred to incubation. The results showed that the best fertilization rate (99.57%) and hatching rate (99.46%) were related to treatment 1 at a concentration of 500 mg / L in 0.5 time It was a minute. Regarding removal of egg adhesion by sodium chloride treatment, the best fertilization rate (87.1) and hatching rate (28.86) were related to 1000 mg / l treatment at 30 minutes. The highest percentage of fertilization (97.52%) and hatching (96.72%) with alkaline enzyme was 50 mg / L treatment at 45 seconds.
